The Human (A2ML1) Alpha-2-Macroglobulin-like Protein 1 ELISA Kit measures Alpha-2-Macroglobulin-Like Protein 1 in samples. The plate has been pre-coated with Human A2ML1 antibody. A2ML1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human A2ML1 Antibody is added and binds to A2ML1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated A2ML1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human A2ML1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a Peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase (By similarity). Displays inhibitory activity against chymotrypsin, papain, thermolysin, subtilisin A and, to a lesser extent, elastase but not trypsin.
