The Human (ACACA) Acetyl-CoA Carboxylase 1 ELISA Kit measures Acetyl-Coa Carboxylase 1 in samples. The plate has been pre-coated with Human ACACA antibody. ACACA present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human ACACA Antibody is added and binds to ACACA in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ACACA antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human ACACA. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Cytosolic enzyme that catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the first and rate-limiting step of de novo fatty acid biosynthesis (PubMed: 20952656, PubMed: 20457939, PubMed: 29899443). This is a 2 steps reaction starting with the ATP-dependent carboxylation of the biotin carried by the biotin carboxyl carrier (BCC) domain followed by the transfer of the carboxyl group from carboxylated biotin to acetyl-CoA (PubMed: 20952656, PubMed: 20457939, PubMed: 29899443).
