The Human (ACO1) Cytoplasmic Aconitate Hydratase ELISA Kit measures Cytoplasmic Aconitate Hydratase in samples. The plate has been pre-coated with Human ACO1 antibody. ACO1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human ACO1 Antibody is added and binds to ACO1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ACO1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human ACO1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Iron sensor. Binds a 4Fe-4S cluster and functions as aconitase when cellular iron levels are high. Functions as mRNA binding protein that regulates uptake, sequestration and utilization of iron when cellular iron levels are low. Binds to iron-responsive elements (IRES) in target mRNA species when iron levels are low. Binding of a 4Fe-4S cluster precludes RNA binding.
