The Human (ITBeta4) Integrin Beta4 ELISA Kit measures Integrin beta4 in samples. The plate has been pre-coated with Human ITGB4 antibody. ITGB4 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human ITGB4 Antibody is added and binds to ITGB4 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ITGB4 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human ITGB4. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Integrin alpha-6/beta-4 is a receptor for laminin. Plays a critical structural role in the hemidesmosome of epithelial cells. Is required for the regulation of keratinocyte polarity and motility. ITGA6:ITGB4 binds to NRG1 (via EGF domain) and this binding is essential for NRG1-ERBB signaling (PubMed: 20682778). ITGA6:ITGB4 binds to IGF1 and this binding is essential for IGF1 signaling (PubMed: 22351760). ITGA6:ITGB4 binds to IGF2 and this binding is essential for IGF2 signaling (PubMed: 28873464).
