The Human (MDH2) Malate Dehydrogenase ELISA Kit measures Malate dehydrogenase in samples. The plate has been pre-coated with Human MDH2 antibody. MDH2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human MDH2 Antibody is added and binds to MDH2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated MDH2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human MDH2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background Malate dehydrogenase, mitochondrial also known as malate dehydrogenase 2 is an enzyme that in humans is encoded by the MDH2 gene. Malate dehydrogenase catalyzes the reversible oxidation of malate to oxaloacetate, utilizing the NAD/NADH cofactor system in the citric acid cycle. The protein encoded by this gene is localized to the mitochondria and may play pivotal roles in the malate-aspartate shuttle that operates in the metabolic coordination between cytosol and mitochondria. Because malate dehydrogenase is closely tied to the citric acid cycle, regulation is highly dependent on TCA products.[8] Citrate also affects MDH activity by very complex manner. It inhibits the reduction of oxaloacetate under all conditions. Citrate also inhibits malate oxidation, but only at low malate or NAD concentrations. Source: UniProt Consortium (2025)
