The Human (MDM2) E3 Ubiquitin-protein Ligase Mdm2 ELISA Kit measures E3 ubiquitin-protein ligase Mdm2 in samples. The plate has been pre-coated with Human MDM2 antibody. MDM2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human MDM2 Antibody is added and binds to MDM2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated MDM2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human MDM2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Mouse double minute 2 homolog (MDM2) also known as E3 ubiquitin-protein ligase Mdm2 is a protein that in humans is encoded by the MDM2 gene. Mdm2 is an important negative regulator of the p53 tumor suppressor. Mdm2 protein functions both as an E3 ubiquitin ligase that recognizes the N-terminal trans-activation domain (TAD) of the p53 tumor suppressor and as an inhibitor of p53 transcriptional activation.
