The Human (METAP2) Methionine Aminopeptidase 2 ELISA Kit measures Methionine Aminopeptidase 2 in samples. The plate has been pre-coated with Human METAP2 antibody. METAP2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human METAP2 Antibody is added and binds to METAP2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated METAP2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human METAP2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Cotranslationally removes the N-terminal methionine from nascent proteins. The N-terminal methionine is often cleaved when the second residue in the primary sequence is small and uncharged (Met-Ala-, Cys, Gly, Pro, Ser, Thr, or Val). The catalytic activity of human METAP2 toward Met-Val Peptides is consistently two orders of magnitude higher than that of METAP1, suggesting that it is responsible for processing proteins containing N-terminal Met-Val and Met-Thr sequences in vivo. Protects eukaryotic initiation factor EIF2S1 from translation-inhibiting phosphorylation by inhibitory kinases such as EIF2AK2/PKR and EIF2AK1/HCR.
