The Human (MYL3) Myosin Light Chain 3 ELISA Kit measures Myosin Light Chain 3 in samples. The plate has been pre-coated with Human MYL3 antibody. MYL3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human MYL3 Antibody is added and binds to MYL3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated MYL3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human MYL3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Myosin essential light chain (ELC), ventricular/cardiac isoform is a protein that in humans is encoded by the MYL3 gene. This cardiac ventricular/slow skeletal ELC isoform is distinct from that expressed in fast skeletal muscle (MYL1) and cardiac atrial muscle (MYL4). Ventricular ELC is part of the myosin molecule and is important in modulating cardiac muscle contraction. Studies have provided evidence for ELC as modulator of myosin crossbridge kinetics. Treating cardiac myofibrils with the lysine-rich N-terminal peptide (amino acids 5-14) evoked a supramaximal increase in cardiac myofibrillar MgATPase activity at submaximal calcium concentrations, and further studies demonstrated that this region of ELC modulates the affinity of myosin for actin.
