The Human (NEDD8) Nedd8 ELISA Kit measures NEDD8 in samples. The plate has been pre-coated with Human NEDD8 antibody. NEDD8 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human NEDD8 Antibody is added and binds to NEDD8 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated NEDD8 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human NEDD8. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Ubiquitin-like protein which plays an important role in cell cycle control and embryogenesis. Covalent attachment to its substrates requires prior activation by the E1 complex UBE1C-APPBP1 and linkage to the E2 enzyme UBE2M. Attachment of NEDD8 to cullins activates their associated E3 ubiquitin ligase activity, and thus promotes polyubiquitination and proteasomal degradation of cyclins and other regulatory proteins.
