The Human (PADI1) Protein-arginine Deiminase Type-1 ELISA Kit measures protein-arginine deiminase type-1 in samples. The plate has been pre-coated with Human PADI1 antibody. PADI1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human PADI1 Antibody is added and binds to PADI1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated PADI1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human PADI1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Peptidyl arginine deiminase, type I, also known as PADI1, is a protein which in humans is encoded by the PADI1 gene. This gene encodes a member of the peptidyl arginine deiminase family of enzymes, which catalyze the post-translational deimination of proteins by converting arginine residues into citrullines in the presence of calcium ions. The family members have distinct substrate specificities and tissue-specific expression patterns. The type I enzyme is involved in the late stages of epidermal differentiation, where it deiminates filaggrin and keratin K1, which maintains hydration of the stratum corneum, and hence the cutaneous barrier function. This enzyme may also play a role in hair follicle formation. This gene exists in a cluster with four other paralogous genes. Source: UniProt Consortium (2025)
