The Human (PADI2) Peptidyl Arginine Deiminase Type-2 ELISA Kit measures Peptidyl Arginine deiminase type-2 in samples. The plate has been pre-coated with Human PADI2 antibody. PADI2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human PADI2 Antibody is added and binds to PADI2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated PADI2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human PADI2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background Protein-arginine deiminase type-2 is an enzyme that in humans is encoded by the PADI2 gene. This gene encodes a member of the peptidyl arginine deiminase family of enzymes, which catalyze the post-translational deimination of proteins by converting arginine residues into citrullines in the presence of calcium ions. The family members have distinct substrate specificities and tissue-specific expression patterns. The type II enzyme is the most widely expressed family member. Known substrates for this enzyme include myelin basic protein in the central nervous system and vimentin in skeletal muscle and macrophages. This enzyme is thought to play a role in the onset and progression of neurodegenerative human disorders, including Alzheimer disease and multiple sclerosis, and it has also been implicated in glaucoma pathogenesis.
