The Human (RECK) Reversion-inducing Cysteine-rich Protein with Kazal Motifs ELISA Kit measures Reversion-Inducing Cysteine-Rich Protein with Kazal Motifs in samples. The plate has been pre-coated with Human RECK antibody. RECK present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human RECK Antibody is added and binds to RECK in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated RECK antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human RECK. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background Functions together with ADGRA2 to enable brain endothelial cells to selectively respond to Wnt7 signals (WNT7A or WNT7B) (PubMed: 28289266, PubMed: 30026314). Plays a key role in Wnt7-specific responses: required for central nervous system (CNS) angiogenesis and blood-brain barrier regulation (By similarity). Acts as a Wnt7-specific coactivator of canonical Wnt signaling by decoding Wnt ligands: acts by interacting specifically with the disordered linker region of Wnt7, thereby conferring ligand selectivity for Wnt7 (PubMed: 30026314). ADGRA2 is then required to deliver RECK-bound Wnt7 to frizzled by assembling a higher-order RECK-ADGRA2-Fzd-LRP5-LRP6 complex (PubMed: 30026314). Also acts as a serine protease inhibitor: negatively regulates Matrix Metalloproteinase-9 (MMP9) by suppressing MMP9 secretion and by direct inhibition of its enzymatic activity (PubMed: 9789069, PubMed: 18194466).
