The Human (BAP1) Ubiquitin Carboxyl Terminalhydrolase Bap1 ELISA Kit measures Ubiquitin carboxyl terminal hydrolase BAP1 in samples. The plate has been pre-coated with Human BAP1 antibody. BAP1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human BAP1 Antibody is added and binds to BAP1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BAP1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human BAP1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
BRCA1 associated protein-1 (ubiquitin carboxy-terminal hydrolase) is a deubiquitinating enzyme that in humans is encoded by the BAP1 gene. BAP1 encodes an 80.4 kDa nuclear-localizing protein with a ubiquitin carboxy-terminal hydrolase (UCH) domain that gives BAP1 its deubiquitinase activity. Recent studies have shown that BAP1 and its fruit fly homolog, Calypso, are members of the polycomb-group proteins (PcG) of highly conserved transcriptional repressors required for long-term silencing of genes that regulate cell fate determination, stem cell pluripotency, and other developmental processes.
