The Human (CAPN5) Calpain-5 ELISA Kit measures Calpain-5 in samples. The plate has been pre-coated with Human CAPN5 antibody. CAPN5 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CAPN5 Antibody is added and binds to CAPN5 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CAPN5 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CAPN5. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Calpain-5 is a protein that in humans is encoded by the CAPN5 gene. Calpains are calcium-dependent cysteine proteases involved in signal transduction in a variety of cellular processes. A functional calpain protein consists of an invariant small subunit and 1 of a family of large subunits. CAPN5 is one of the large subunits. Unlike some of the calpains, CAPN5 and CAPN6 lack a calmodulin-like domain IV. Because of the significant similarity to Caenorhabditis elegans sex determination gene tra-3, CAPN5 is also called as HTRA3. Source: UniProt Consortium (2025)
