The Human (CD209) Cd209 Antigen ELISA Kit measures CD209 antigen in samples. The plate has been pre-coated with Human CD209 antibody. CD209 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CD209 Antibody is added and binds to CD209 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CD209 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CD209. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background DC-SIGN (Dendritic Cell-Specific Intercellular adhesion molecule-3-Grabbing Non-integrin) also known as CD209 (Cluster of Differentiation 209) is a protein which in humans is encoded by the CD209 gene. DC-SIGN is a C-type lectin receptor present on the surface of both macrophages and dendritic cells. DC-SIGN on macrophages recognises and binds with high affinity to high-mannose type N-glycans, a class of PAMPs (pathogen associated molecular patterns) commonly found on viruses, bacteria and fungi. This binding interaction activates phagocytosis. On myeloid and pre-plasmacytoid dendritic cells DC-SIGN mediates dendritic cell rolling interactions with blood endothelium and activation of CD4+ T cells, as well as recognition of pathogen haptens.
