The Human (CDKN1A) Cyclin-dependent Kinase Inhibitor 1 ELISA Kit measures Cyclin-Dependent Kinase Inhibitor 1 in samples. The plate has been pre-coated with Human CDKN1A antibody. CDKN1A present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CDKN1A Antibody is added and binds to CDKN1A in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CDKN1A antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CDKN1A. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
May be involved in p53/TP53 mediated inhibition of cellular proliferation in response to DNA damage. Binds to and inhibits cyclin-dependent kinase activity, preventing phosphorylation of critical cyclin-dependent kinase substrates and blocking cell cycle progression. Functions in the nuclear localization and assembly of cyclin D-CDK4 complex and promotes its kinase activity towards RB1. At higher stoichiometric ratios, inhibits the kinase activity of the cyclin D-CDK4 complex. Inhibits DNA synthesis by DNA polymerase delta by competing with POLD3 for PCNA binding (PubMed: 11595739).
