The Human (CLSPN) Claspin ELISA Kit measures Claspin in samples. The plate has been pre-coated with Human CLSPN antibody. CLSPN present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CLSPN Antibody is added and binds to CLSPN in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CLSPN antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CLSPN. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Required for checkpoint mediated cell cycle arrest in response to inhibition of DNA replication or to DNA damage induced by both ionizing and UV irradiation. Adapter protein which binds to BRCA1 and the checkpoint kinase CHEK1 and facilitates the ATR-dependent phosphorylation of both proteins. Can also bind specifically to branched DNA structures and may associate with S-phase chromatin following formation of the pre-replication complex (pre-RC).
