The Human (CST1) Cystatin-Sn ELISA Kit measures Cystatin-Sn in samples. The plate has been pre-coated with Human CST1 antibody. CST1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CST1 Antibody is added and binds to CST1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CST1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CST1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Human saliva appears to contain several cysteine proteinase inhibitors that are immunologically related to cystatin S but that differ in their specificity due to amino acid sequence differences. Cystatin SN, with a pI of 7.5, is a much better inhibitor of papain and dipeptidyl peptidase I than is cystatin S, although both inhibit ficin equally well. Source: UniProt Consortium (2025)
