The Human (DKK1) Dickkopf-1 ELISA Kit measures Dickkopf-1 in samples. The plate has been pre-coated with Human DKK1 antibody. DKK1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human DKK1 Antibody is added and binds to DKK1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated DKK1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human DKK1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6 (PubMed: 22000856). DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease (PubMed: 17143291). Inhibits the pro-apoptotic function of KREMEN1 in a Wnt-independent manner, and has anti-apoptotic activity (By similarity). Source: UniProt Consortium (2025)
