The Human (DLG4) Disks Large Homolog 4 ELISA Kit measures Disks Large Homolog 4 in samples. The plate has been pre-coated with Human DLG4 antibody. DLG4 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human DLG4 Antibody is added and binds to DLG4 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated DLG4 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human DLG4. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Postsynaptic scaffolding protein that plays a critical role in synaptogenesis and synaptic plasticity by providing a platform for the postsynaptic clustering of crucial synaptic proteins. Interacts with the cytoplasmic tail of NMDA receptor subunits and shaker-type potassium Channels. Required for synaptic plasticity associated with NMDA receptor signaling. Overexpression or depletion of DLG4 changes the ratio of excitatory to inhibitory synapses in hippocampal neurons. May reduce the amplitude of ASIC3 acid-evoked currents by retaining the Channel intracellularly. May regulate the intracellular trafficking of ADR1B. Also regulates AMPA-type glutamate receptor (AMPAR) immobilization at postsynaptic density keeping the Channels in an activated state in the presence of glutamate and preventing synaptic depression.
