The Human (ERAP1) Endoplasmic Reticulum Aminopeptidase 1 ELISA Kit measures Endoplasmic Reticulum Aminopeptidase 1 in samples. The plate has been pre-coated with Human ERAP1 antibody. ERAP1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human ERAP1 Antibody is added and binds to ERAP1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ERAP1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human ERAP1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Aminopeptidase that plays a central role in Peptide trimming, a step required for the generation of most HLA class I-binding Peptides. Peptide trimming is essential to customize longer precursor Peptides to fit them to the correct length required for presentation on MHC class I molecules. Strongly prefers substrates 9-16 residues long. Rapidly degrades 13-mer to a 9-mer and then stops. Preferentially hydrolyzes the residue Leu and Peptides with a hydrophobic C-terminus, while it has weak activity toward Peptides with charged C-terminus. May play a role in the inactivation of Peptide hormones.
