The Human (FZD10) Frizzled-10 ELISA Kit measures Frizzled-10 in samples. The plate has been pre-coated with Human FZD10 antibody. FZD10 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human FZD10 Antibody is added and binds to FZD10 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated FZD10 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human FZD10. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

The canonical Wnt/beta-catenin signaling pathway leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes. A second signaling pathway involving PKC and calcium fluxes has been seen for some family members, but it is not yet clear if it represents a distinct pathway or if it can be integrated in the canonical pathway, as PKC seems to be required for Wnt-mediated inactivation of GSK-3 kinase. Both pathways seem to involve interactions with G-proteins.
