The Human (GPA33) Cell Surface A33 Antigen ELISA Kit measures Cell surface A33 antigen in samples. The plate has been pre-coated with Human GPA33 antibody. GPA33 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human GPA33 Antibody is added and binds to GPA33 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GPA33 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human GPA33. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Cell surface A33 antigen is a protein that in humans is encoded by the GPA33 gene. The glycoprotein encoded by this gene is a cell surface antigen that is expressed in greater than 95% of human colon cancers. The open reading frame encodes a 319-amino acid polypeptide having a putative secretory signal sequence and 3 potential glycosylation sites. The predicted mature protein has a 213-amino acid extracellular region, a single transmembrane domain, and a 62-amino acid intracellular tail. The sequence of the extracellular region contains 2 domains characteristic of the CD2 subgroup of the immunoglobulin (Ig) superfamily. Source: UniProt Consortium (2025)
