Human GZMS-B (Granzymes B) ELISA Kit
The Human (GZMS-B) Granzymes B ELISA Kit measures Granzymes B in samples. The plate has been pre-coated with Human GZMB antibody. GZMB present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human GZMB Antibody is added and binds to GZMB in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GZMB antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human GZMB. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Abundant protease in the cytosolic granules of cytotoxic T-cells and NK-cells which activates caspase-independent pyroptosis when delivered into the target cell through the immunological synapse (PubMed: 3262682, PubMed: 3263427, PubMed: 1985927). It cleaves after Asp (PubMed: 8258716, PubMed: 1985927). Once delivered into the target cell, acts by catalyzing cleavage of gasdermin-E (GSDME), releasing the pore-forming moiety of GSDME, thereby triggering pyroptosis and target cell death (PubMed: 32188940, PubMed: 31953257). Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution.
