The Human (RSPO2) R-Spondin-2 ELISA Kit measures R-Spondin-2 in samples. The plate has been pre-coated with Human RSPO2 antibody. RSPO2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human RSPO2 Antibody is added and binds to RSPO2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated RSPO2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human RSPO2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Activator of the canonical Wnt signaling pathway by acting as a ligand for LGR4-6 receptors. Upon binding to LGR4-6 (LGR4, LGR5 or LGR6), LGR4-6 associate with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. Also regulates the canonical Wnt/beta-catenin-dependent pathway and non-canonical Wnt signaling by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway (PubMed: 21909076, PubMed: 21727895, PubMed: 22615920). During embryonic development, plays a crucial role in limb specification, amplifying the Wnt signaling pathway independently of LGR4-6 receptors, possibly by acting as a direct antagonistic ligand to RNF43 and ZNRF3, hence governing the number of limbs an embryo should form (PubMed: 29769720).
