The Human (SHANK3) Sh3 and Multiple Ankyrin Repeat Domains Protein 3 ELISA Kit measures SH3 and multiple ankyrin repeat domains protein 3 in samples. The plate has been pre-coated with Human SHANK3 antibody. SHANK3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human SHANK3 Antibody is added and binds to SHANK3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated SHANK3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human SHANK3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background Major scaffold postsynaptic density protein which interacts with multiple proteins and complexes to orchestrate the dendritic spine and synapse formation, maturation and maintenance. Interconnects receptors of the postsynaptic membrane including NMDA-type and metabotropic glutamate receptors via complexes with GKAP/PSD-95 and HOMER, respectively, and the actin-based cytoskeleton. Plays a role in the structural and functional organization of the dendritic spine and synaptic junction through the interaction with Arp2/3 and WAVE1 complex as well as the promotion of the F-actin clusters. By way of this control of actin dynamics, participates in the regulation of developing neurons growth cone motility and the NMDA receptor-signaling. Also modulates GRIA1 exocytosis and GRM5/MGLUR5 expression and signaling to control the AMPA and metabotropic glutamate receptor-mediated synaptic transmission and plasticity.
