The Human (SMO) Smoothened Homolog ELISA Kit measures Smoothened Homolog in samples. The plate has been pre-coated with Human SMO antibody. SMO present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human SMO Antibody is added and binds to SMO in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated SMO antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human SMO. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background G protein-coupled receptor that probably associates with the patched protein (PTCH) to transduce the hedgehog's proteins signal. Binding of sonic hedgehog (SHH) to its receptor patched is thought to prevent normal inhibition by patched of smoothened (SMO). Required for the accumulation of KIF7, GLI2 and GLI3 in the cilia (PubMed: 19592253).
