The Human (SMYD2) N-lysine Methyltransferase Smyd2 ELISA Kit measures N-lysine methyltransferase SMYD2 in samples. The plate has been pre-coated with Human SMYD2 antibody. SMYD2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human SMYD2 Antibody is added and binds to SMYD2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated SMYD2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human SMYD2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Protein-lysine N-methyltransferase that methylates both histones and non-histone proteins, including p53/TP53 and RB1. Specifically trimethylates histone H3 'Lys-4' (H3K4me3) in vivo. The activity requires interaction with HSP90alpha. Shows even higher methyltransferase activity on p53/TP53. Monomethylates 'Lys-370' of p53/TP53, leading to decreased DNA-binding activity and subsequent transcriptional regulation activity of p53/TP53. Monomethylates RB1 at 'Lys-860'.
