The Human (SPAM1) Hyaluronidase Ph-20 ELISA Kit measures Hyaluronidase Ph-20 in samples. The plate has been pre-coated with Human SPAM1 antibody. SPAM1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human SPAM1 Antibody is added and binds to SPAM1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated SPAM1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human SPAM1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Involved in sperm-egg adhesion. Upon fertilization sperm must first penetrate a layer of cumulus cells that surrounds the egg before reaching the zona pellucida. The cumulus cells are embedded in a matrix containing hyaluronic acid which is formed prior to ovulation. This protein aids in penetrating the layer of cumulus cells by digesting hyaluronic acid.
