The Human (TM) Thrombomodulin ELISA Kit measures Thrombomodulin in samples. The plate has been pre-coated with Human THBD antibody. THBD present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human THBD Antibody is added and binds to THBD in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated THBD antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human THBD. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Thrombomodulin is a specific endothelial cell receptor that forms a 1:1 stoichiometric complex with thrombin. This complex is responsible for the conversion of protein C to the activated protein C (protein Ca). Once evolved, protein Ca scissions the activated cofactors of the coagulation mechanism, factor Va and factor VIIIa, and thereby reduces the amount of thrombin generated.
