The Human (TSPAN8) Tetraspanin-8 ELISA Kit measures Tetraspanin-8 in samples. The plate has been pre-coated with Human TSPAN8 antibody. TSPAN8 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human TSPAN8 Antibody is added and binds to TSPAN8 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated TSPAN8 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human TSPAN8. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Tetraspanin-8 is a protein that in humans is encoded by the TSPAN8 gene. The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein that is known to complex with integrins. This gene is expressed in different carcinomas.
