The Human (TUBB3) Tubulin Beta-3 Chain ELISA Kit measures Tubulin Beta-3 Chain in samples. The plate has been pre-coated with Human TUBB3 antibody. TUBB3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human TUBB3 Antibody is added and binds to TUBB3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated TUBB3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human TUBB3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Tubulin is the major constituent of microtubules. It binds two moles of GTP, one at an exchangeable site on the beta chain and one at a non-exchangeable site on the alpha chain. TUBB3 plays a critical role in proper axon guidance and mantainance. Binding of NTN1/Netrin-1 to its receptor UNC5C might cause dissociation of UNC5C from polymerized TUBB3 in microtubules and thereby lead to increased microtubule dynamics and axon repulsion (PubMed: 28483977).
