The Mouse (ATG4A) Autophagy Related Protein 4A ELISA Kit measures Autophagy related protein 4A in Mouse samples. The plate has been pre-coated with Mouse ATG4A antibody. ATG4A present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse ATG4A Antibody is added and binds to ATG4A in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ATG4A antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse ATG4A. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Cysteine protease required for the cytoplasm to vacuole transport (Cvt) and autophagy. Cleaves the C-terminal amino acid of ATG8 family proteins to reveal a C-terminal glycine. Exposure of the glycine at the C-terminus is essential for ATG8 proteins conjugation to phosphatidylethanolamine (PE) and insertion to membranes, which is necessary for autophagy. Preferred substrate is GABARAPL2 followed by MAP1LC3A and GABARAP. Has also an activity of delipidating enzyme for the PE-conjugated forms (By similarity). Source: UniProt Consortium (2025)
