The Mouse (BMI-1) Polycomb Complex Protein Bmi-1 ELISA Kit measures Polycomb complex protein BMI-1 in Mouse samples. The plate has been pre-coated with Mouse BMI1 antibody. BMI1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse BMI1 Antibody is added and binds to BMI1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BMI1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse BMI1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. The complex composed of RNF2, UB2D3 and BMI1 binds nucleosomes, and has activity only with nucleosomal histone H2A.
