The Mouse (CASP3) Caspase 3 ELISA Kit measures Caspase 3 in Mouse samples. The plate has been pre-coated with Mouse CASP3 antibody. CASP3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse CASP3 Antibody is added and binds to CASP3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CASP3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse CASP3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Involved in the activation cascade of caspases responsible for apoptosis execution. At the onset of apoptosis it proteolytically cleaves poly(ADP-ribose) polymerase (PARP) at a '216-Asp-|-Gly-217' bond. Cleaves and activates sterol regulatory element binding proteins (SREBPs) between the basic helix-loop-helix leucine zipper domain and the membrane attachment domain. Cleaves and activates caspase-6, -7 and -9. Triggers cell adhesion in sympathetic neurons through RET cleavage (By similarity). Cleaves IL-1 beta between an Asp and an Ala, releasing the mature cytokine which is involved in a variety of inflammatory processes (By similarity). Cleaves and inhibits serine/threonine-protein kinase AKT1 in response to oxidative stress (PubMed: 12124386). Source: UniProt Consortium (2025)
