The Mouse (HIF-1A) Hypoxia-inducible Factor 1Α ELISA Kit measures Hypoxia-Inducible Factor 1 alpha in Mouse samples. The plate has been pre-coated with Mouse HIF1A antibody. HIF1A present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse HIF1A Antibody is added and binds to HIF1A in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated HIF1A antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse HIF1A. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background Functions as a master transcriptional regulator of the adaptive response to hypoxia. Under hypoxic conditions, activates the transcription of over 40 genes, including erythropoietin, glucose transporters, glycolytic enzymes, vascular endothelial growth factor, HILPDA, and other genes whose protein products increase oxygen delivery or facilitate metabolic adaptation to hypoxia. Plays an essential role in embryonic vascularization, tumor angiogenesis and pathophysiology of ischemic disease. Heterodimerizes with ARNT; heterodimer binds to core DNA sequence 5'-TACGTG-3' within the hypoxia response element (HRE) of target gene promoters (PubMed: 26245371). Activation requires recruitment of transcriptional coactivators such as CREBBP and EP300. Activity is enhanced by interaction with both, NCOA1 or NCOA2. Interaction with redox regulatory protein APEX seems to activate CTAD and potentiates activation by NCOA1 and CREBBP.
