Mouse PAH (Phenylalanine-4-hydroxylase) ELISA Kit
The Mouse (PAH) Phenylalanine-4-hydroxylase ELISA Kit measures Phenylalanine-4-hydroxylase in Mouse samples. The plate has been pre-coated with Mouse PAH antibody. PAH present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse PAH Antibody is added and binds to PAH in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated PAH antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse PAH. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Phenylalanine hydroxylase (PAH) (EC 1.14.16.1) is an enzyme that catalyzes the hydroxylation of the aromatic side-chain of phenylalanine to generate tyrosine. PAH is one of three members of the biopterin-dependent aromatic amino acid hydroxylases, a class of monooxygenase that uses tetrahydrobiopterin (BH4, a pteridine cofactor) and a non-heme iron for catalysis. During the reaction, molecular oxygen is heterolytically cleaved with sequential incorporation of one oxygen atom into BH4 and phenylalanine substrate. In humans, mutations in its encoding gene, PAH, can lead to the metabolic disorder phenylketonuria. Source: UniProt Consortium (2025)
