The Mouse (WFDC2) Wap Four-Disulfide Core Domain Protein 2 ELISA Kit measures WAP four-disulfide core domain protein 2 in Mouse samples. The plate has been pre-coated with Mouse WFDC2 antibody. WFDC2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Mouse WFDC2 Antibody is added and binds to WFDC2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated WFDC2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Mouse WFDC2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
WAP four-disulfide core domain protein 2 - also known as Human Epididymis Protein 4 (HE4) - is a protein that in humans is encoded by the WFDC2 gene. HE4 is a tumor marker of ovarian cancer, with 80% sensitivity at a cut-off of 150 pmol/L. This gene encodes a protein that is a member of the WFDC domain family. The WFDC domain, or WAP Signature motif, contains eight cysteines forming four disulfide bonds at the core of the protein, and functions as a protease inhibitor in many family members. This gene is expressed in pulmonary epithelial cells, and was also found to be expressed in some ovarian cancers.[5] The encoded protein is a small secretory protein, which may be involved in sperm maturation.
