The Rat (ALPHA-1-M) Alpha-1-macroglobulin ELISA Kit measures Alpha-1-Macroglobulin in Rat samples. The plate has been pre-coated with Rat A1M antibody. A1M present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat A1M Antibody is added and binds to A1M in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated A1M antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat A1M. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a Peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase (By similarity). Source: UniProt Consortium (2025)
