The Rat (CYP21A2) Steroid 21-hydroxylase ELISA Kit measures Steroid 21-hydroxylase in Rat samples. The plate has been pre-coated with Rat CYP21A2 antibody. CYP21A2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat CYP21A2 Antibody is added and binds to CYP21A2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CYP21A2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat CYP21A2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
A cytochrome P450 monooxygenase that plays a major role in adrenal steroidogenesis. Catalyzes the hydroxylation at C-21 of progesterone and 17alpha-hydroxyprogesterone to respectively form 11-deoxycorticosterone and 11-deoxycortisol, intermediate metabolites in the biosynthetic pathway of mineralocorticoids and glucocorticoids. Mechanistically, uses molecular oxygen inserting one oxygen atom into a substrate, and reducing the second into a water molecule, with two electrons provided by NADPH via cytochrome P450 reductase (CPR; NADPH-ferrihemoprotein reductase).
