The Rat (ET-3) Endothelin 3 ELISA Kit measures Endothelin 3 in Rat samples. The plate has been pre-coated with Rat ET-3 antibody. ET-3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat ET-3 Antibody is added and binds to ET-3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated ET-3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat ET-3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Endothelin-3 is a protein that in humans is encoded by the EDN3 gene. The protein encoded by this gene is a member of the endothelin family. Endothelins are endothelium-derived vasoactive peptides involved in a variety of biological functions. The active form of this protein is a 21 amino acid peptide processed from the precursor protein. The active peptide is a ligand for endothelin receptor type B (EDNRB). The interaction of this endothelin with EDNRB is essential for development of neural crest-derived cell lineages, such as melanocytes and enteric neurons. Mutations in this gene and EDNRB have been associated with Hirschsprung disease (HSCR) and Waardenburg syndrome (WS), which are congenital disorders involving neural crest-derived cells.
