The Rat (GAD2) Glutamate Decarboxylase 2 ELISA Kit measures Glutamate decarboxylase 2 in Rat samples. The plate has been pre-coated with Rat GAD2 antibody. GAD2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat GAD2 Antibody is added and binds to GAD2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GAD2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat GAD2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Glutamate decarboxylase 2 is an enzyme that in humans is encoded by the GAD2 gene. This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantibody and an autoreactive T cell target in insulin-dependent diabetes.
