The Rat (GLUT4) Glucose Transporter 4 ELISA Kit measures Glucose Transporter 4 in Rat samples. The plate has been pre-coated with Rat SLC2A4 antibody. SLC2A4 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat SLC2A4 Antibody is added and binds to SLC2A4 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated SLC2A4 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat SLC2A4. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Insulin-regulated facilitative glucose transporter, which plays a key role in removal of glucose from circulation (PubMed: 2649253, PubMed: 2645527, PubMed: 2211693). Response to insulin is regulated by its intracellular localization: in the absence of insulin, it is efficiently retained intracellularly within storage compartments in muscle and fat cells (PubMed: 2649253, PubMed: 2645527, PubMed: 2211693). Upon insulin stimulation, translocates from these compartments to the cell surface where it transports glucose from the extracellular milieu into the cell (PubMed: 2649253, PubMed: 2645527, PubMed: 2211693).
