The Rat (GRIN2B) Glutamate[NMDA] Receptor Subunit Epsilon-2 ELISA Kit measures Glutamate[Nmda]Receptor Subunit Epsilon-2 in Rat samples. The plate has been pre-coated with Rat GRIN2B antibody. GRIN2B present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat GRIN2B Antibody is added and binds to GRIN2B in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GRIN2B antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat GRIN2B. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Component of NMDA receptor complexes that function as heterotetrameric, ligand-gated ion Channels with high calcium permeability and voltage-dependent sensitivity to magnesium. Channel activation requires binding of the neurotransmitter glutamate to the epsilon subunit, glycine binding to the zeta subunit, plus membrane depolarization to eliminate Channel inhibition by Mg(2+) (PubMed: 1350383, PubMed: 19910922, PubMed: 21677647, PubMed: 24876489, PubMed: 27135925, PubMed: 27916457). Sensitivity to glutamate and Channel kinetics depend on the subunit composition (Probable). In concert with DAPK1 at extrasynaptic sites, acts as a central mediator for stroke damage. Its phosphorylation at Ser-1303 by DAPK1 enhances synaptic NMDA receptor Channel activity inducing injurious Ca2+ influx through them, resulting in an irreversible neuronal death. Contributes to neural pattern formation in the developing brain. Plays a role in long-term depression (LTD) of hippocampus membrane currents and in synaptic plasticity (By similarity).
