The Rat (HPT) Haptoglobin ELISA Kit measures Haptoglobin in Rat samples. The plate has been pre-coated with Rat HP antibody. HP present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat HP Antibody is added and binds to HP in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated HP antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat HP. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
As a result of hemolysis, hemoglobin is found to accumulate in the kidney and is secreted in the urine. Haptoglobin captures, and combines with free plasma hemoglobin to allow hepatic recycling of heme iron and to prevent kidney damage. Haptoglobin also acts as an antioxidant, has antibacterial activity and plays a role in modulating many aspects of the acute phase response. Hemoglobin/haptoglobin complexes are rapidly cleared by the macrophage CD163 scavenger receptor expressed on the surface of liver Kupfer cells through an endocytic lysosomal degradation pathway (By similarity).
