The Rat (KL) Klotho ELISA Kit measures Klotho in Rat samples. The plate has been pre-coated with Rat KL antibody. KL present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat KL Antibody is added and binds to KL in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated KL antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat KL. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
May have weak glycosidase activity towards glucuronylated steroids. However, it lacks essential active site Glu residues at positions 241 and 874, suggesting it may be inactive as a glycosidase in vivo. May be involved in the regulation of calcium and phosphorus homeostasis by inhibiting the synthesis of active vitamin D (By similarity). Essential factor for the specific interaction between FGF23 and FGFR1 (By similarity). The Klotho Peptide generated by cleavage of the membrane-bound isoform may be an anti-aging circulating hormone which would extend life span by inhibiting insulin/IGF1 signaling.
