The Rat (PRNP) Major Prion Protein ELISA Kit measures Major Prion Protein in Rat samples. The plate has been pre-coated with Rat PRNP antibody. PRNP present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat PRNP Antibody is added and binds to PRNP in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated PRNP antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat PRNP. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Its primary physiological function is unclear. May play a role in neuronal development and synaptic plasticity. May be required for neuronal myelin sheath maintenance. May promote myelin homeostasis through acting as an agonist for ADGRG6 receptor. May play a role in iron uptake and iron homeostasis. Soluble oligomers are toxic to cultured neuroblastoma cells and induce apoptosis (in vitro) (By similarity). Association with GPC1 (via its heparan sulfate chains) targets PRNP to lipid rafts.
