The Rat (RUNX2) Runt-related Transcription Factor 2 ELISA Kit measures Runt-related transcription factor 2 in Rat samples. The plate has been pre-coated with Rat RUNX2 antibody. RUNX2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat RUNX2 Antibody is added and binds to RUNX2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated RUNX2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat RUNX2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Transcription factor involved in osteoblastic differentiation and skeletal morphogenesis. Essential for the maturation of osteoblasts and both intramembranous and endochondral ossification. CBF binds to the core site, 5'-PYGPYGGT-3', of a number of enhancers and promoters, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, osteocalcin, osteopontin, bone sialoprotein, alpha 1(I) collagen, LCK, IL-3 and GM-CSF promoters. Inhibits KAT6B-dependent transcriptional activation. In osteoblasts, supports transcription activation: synergizes with SPEN/MINT to enhance FGFR2-mediated activation of the osteocalcin FGF-responsive element (OCFRE). Source: UniProt Consortium (2025)
