The Rat (UCP1) Mitochondrial Brown Fat Uncoupling Protein 1 ELISA Kit measures Mitochondrial Brown Fat Uncoupling Protein 1 in Rat samples. The plate has been pre-coated with Rat UCP1 antibody. UCP1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat UCP1 Antibody is added and binds to UCP1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated UCP1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat UCP1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Background
Mitochondrial protein responsible for thermogenic respiration, a specialized capacity of brown adipose tissue and beige fat that participates in non-shivering adaptive thermogenesis to temperature and diet variations and more generally to the regulation of energy balance (By similarity). Functions as a long-chain fatty acid/LCFA and proton symporter, simultaneously transporting one LCFA and one proton through the inner mitochondrial membrane. However, LCFAs remaining associated with the transporter via their hydrophobic tails, it results in an apparent transport of protons activated by LCFAs (PubMed: 12479871, PubMed: 16814247). Thereby, dissipates the mitochondrial proton gradient and converts the energy of substrate oxydation into heat instead of ATP.
